do a shetload of megatian quests as they appear. eventually you should be able to access that tunnel.

There's no "boss one"- the Boss is in a normal, easily accessible "Hidden Street" map. The two you're probably asking about are either the door at the far right of the town map with all the NPCs you have to do quests for, or the place you have to steal the jewels from. And you're probably asking about the first one. The answer is, you CAN'T go in there until you finish doing ALL the quests for the three NPCs who give you "tests" to see if you're worthy of becoming a Sand Bandit. Once you finish all their quests you can enter the secret door just by pressing "up" normally.
